Absorption Spectra and Reaction with Haptoglobin of Hemoglobin (a-NO, PUnliganded)

چکیده

Human hemoglobin (aNOD), in which a! chains were liganded with nitric oxide and p chains were unliganded, showed a characteristic spectrum in the Soret region. Its absorption was significantly reduced compared with the averaged absorption of IX-NO and p. On the other hand, a complex of (cuNO0) with haptoglobin did not show such a characteristic spectrum. It was concluded, therefore, that there are some subunit interactions in (cYNOP) and that they are lost when (otNO@ is bound to haptoglobin. Reaction of (CXNOP) with haptoglobin was followed spectrophotometrically, and it was found that the rate of reaction of (aNO/?) with haptoglobin was markedly different from that of fully liganded or fully unliganded hemoglobin with haptoglobin. (olNO0) seems to be in a different conformational state from both fully liganded or fully unliganded hemoglobin.

Differences in Spectra of a and p Chains of Hemoglobin between Isolated State and in Tetramer

The absorption spectrum of deoxygenated human hemoglobin differs from that of the isolated a! and /3 chains, and the difference has been ascribed to the conformational changes on which depends the cooperative ligand binding. Studies on hybrid-heme hemoglobins, in which one chain contains protoheme and the other chain mesoheme, showed that the prominent peak in the Soret region of the difference...

The binding of hemoglobin to haptoglobin and its relation to subunit dissociation of hemoglobin.

The binding of liganded hemoglobin to haptoglobin, a plasma cr2-glycoprotein, is an irreversible and stoichiometric reaction that occurs physiologically at the micromolar concentration level. The study of the reaction between deoxyhemoglobin which does not bind haptoglobin and a haptoglobin solution saturated with carbon monoxide indicates that hemoglobin tetramer is incapable of binding haptog...

A new method for Eosin preparation and examination of its absorption and emission spectra

Eosin is one of the original dyes of xanthenes derivatives. It is also one of the main derivatives of fluorescein and has brilliant fluorescence. Eosin disodium salt called "Eosin Y" is the most useful form of this compound. In common procedures, eosin is usually obtained by the action of bromine on fluorescein, which has not good yield and purity degree. In this research, it is prepared by the...

Heterotropic Effect of β-lactam Antibiotics on Antioxidant Property of Haptoglobin) 2-2(-Hemoglobin Complex

Haptoglobin (Hp) is a mammalian serum glycoprotein showing a genetic polymorphism with three types, 1-1, 2-2 and 1-2. Hp appears to conserve the recycling of heme-iron by forming an essentially irreversible but non-covalent complex with hemoglobin which is released into the plasma by erythrocyte lysis. As an important consequence, Haptoglobin-Hemoglobin complex (Hp-Hb) shows considerable antiox...
